Abstract

Objective To explore the effect of Emodin on expression of vascular endothelial growth factor (VEGF) and nuclear factor-kappa B (NF-κB) in lung tissue of lipopolysaccharide induced acute lung injury (ALI) rats. Methods Male rats were divided into model group, Emodin group, saline group and normal control group, 10 rats in each group. ALI rats were modeled in model group, Emodin group and saline group. The control group was given conventional feeding; Emodin group and saline group were given 20 mg/kg Emodin and normal saline respectively 1 h before ALI establishment. Rats were sacrificed 4 h after modeling. The morphological characteristics was observed by HE staining. Levels of serum TNF-a and IL-6 were detected by ELISA. Immunohistochemical staining, real-time PCR and Western blot were utilized to test the expressions of VEGF and NF-kB. Results The lung tissue structure of control group was clear and there was no inflammatory cell infiltration. The lung tissue structure in model group and saline group was accompanied by a lot of inflammatory cell infiltration. The lung tissue structure of Emodin group was unclear and there was a small amount of inflammatory cell infiltration. Compared with the control group, the expression of VEGF (F = 40.392 and 40.019, all P = 0.000), NF-κB (F = 45.027 and 37.978,all P = 0.000), VEGF mRNA and NF-κB mRNA in model group and saline group were significantly increased (P < 0.05), W/D was increased and serum TNF-a and IL-6 were significantly increased (F= 3.477, 54.757, 144.139, 62.795, 29.924, and 156.352, all P = 0.000). The levels ofVEGF (F = 8.036, P = 0.011), NF-κB (F = 8.728, P = 0.008), VEGF mRNA and NF-κB mRNA in Emodin group were significantly decreased (P < 0.05); W/D, serum TNF-a and IL-6 were significantly decreased (F = 10.453, 54.198, and 75.777, P = 0.005, 0.000 and 0.000). Conclusion Emodin can reduce pulmonary edema and relieve lung injury degree in ALI, of which the mechanism is related to down-regulation of VEGF and NF-κB.
