摘要

Abstract

Objective To investigate the effect of anesthesia on Sprague-Dawley (SD) rats model of severe seawater-immersed hypothermia. Methods Forty male SD rats, which were implanted intraperitoneally with temperature recorder 10 days before the experiment, were randomly divided into two groups: the control group (n = 20, injected intraperitoneal with physiological saline) and the anesthesia group (n = 20, injected intraperitoneal with 1.5% pentobarbital sodium). Ten minutes after intraperitoneal injection, the rats of the two groups were given 15 ℃ seawater immersion for 5 h, and the status of rats were recorded, such as life indicators, the condition of activities-awareness, muscle fasciculation, mortality, etc. The rats were sacrificed at the end of the experiment to remove the temperature recorder to obtain the dynamic temperature data. Results After seawater immersion, the intraperitoneal temperature of the control group and the anesthesia group decreased gradually, and in about 2 h the temperature were close to the water temperature respectively. The temperature of the anesthesia group decreased faster in the earlier immersing period. In the control group, the level of activity consciousness, heart rate and respiration rate were increased first and then decreased to a low level after 2 h, reaching a peak between the 10 min and 20 min, while the above indexes in the anesthesia group decreased rapidly first and then slowly, and maintained a low level after 2 h immersion. The muscle fasciculation frequency in the two groups both increased first and then decreased, and the peak values of the two groups both appeared at 20 min, and the muscle fasciculation of the control group completely disappeared later than the anesthesia group. The mortality rate in the control group was significantly lower than that in the anesthesia group, and the median death time of the control group was significantly later than that in anesthesia group. Conclusion Anesthesia has a significant effect on consciousness, respiration, heart rate and muscle fasciculation in SD rats with hypothermia seawater immersion, and the mortality of hypothermia rats is significantly increased by anesthesia, and there is an advantage to apply rats model without anesthesia in the study of the interaction between hypothermia and nervous system, the stress response in hypothermia, energy metabolism at low body temperature, and so on.
