Abstract
Objective To study the molecular mechanism of the ABO blood group subtypes in 95 Chinese people.Methods Samples were collected during Nov, 2013-Oct, 2017 which are forward-reverse-typing inconsistent; serological tests were conducted on these samples and genotyping by sequence-specific primer polymerase chain reaction (PCR-SSP) was performed. The 6 th—7 th exon gene was sequenced. Some of the specimens were sequenced through the 1 st—7 th exon. Results A total of 95 cases (73 cases of first certificate and 22 family members) was confirmed by blood type serological method.The PCR-SSP and gene sequencing tests revealed that, in the 73 cases, 13 cases (17. 8%) were subtype A; 17 cases (23. 3%) were subtype B; 22 cases (30. 1%) were type B (A)-type, yielding a total detection rate of 71. 2% (52/73). 18 cases of ABO subtype gene were detected in the 22 pedigree members. 21 of the 73 cases (28. 8%, 21/73) failed in ABO typing. 5 of the 21 cases (6. 8%, 5/73) was detected with O gene but expressed weak A or weak B antigen; In 4 cases of 22 family members, the ABO subtype gene was not detected, and a total of 25 cases (25/95) were reported. Conclusion The genotypes and phenotypes of ABO subtypes did not completely match, and the same genotype of blood group serotype corresponded to different genotypes, and the same genotype showed different serological results.关键词
