摘要

Abstract

Objective To assess the stability of HCV-RNA detection on voluntary blood donor samples at various storage conditions. Methods We studied a viral positive sample with a HCV-RNA concentration lower than 102 IU/m L. The Sample was subjected to the following conditions: 1) divided the sample into two tubes (50 m L), and 2) one tube was stored at4℃ for 4 h, 24 h, 48 h, 72, 7 d, 14 d, 3) the other was stored at-20℃ and went through one freeze-thaw cycle at the same time, 4) repeated quantitative testing HCV-RNA by Roche Cobas s 201 Config D 5 times for one sample at each test. Results almost all tests reported positive; the quantitative test result showed that samples stored at-20℃ and freeze-thawed presented decreased HCV-RNA readings as time increased. Reading values (IU/m L) at 4/24/48/72 h/7/14 d are as follows:36. 9; 62. 6, 89. 4, 58. 8, 88. 1, 68. 8; 68. 0, 67. 6, 40. 2, 57. 9, 61. 6; <15. 0, 19. 3, 69. 9, 37. 5, 56. 8; 16. 9, 21. 6, < 15. 0, 37. 7, <15. 0; 26. 4, 30. 8, 49. 4, <15. 0, 42. 7. One sample went below the detection limit (<15. 0) on the 4 th, 5 th and 6 th rounds of the freeze-thaw cycle. The HCV-RNA concentration in the 4℃ group had not changed much, but Nucleic Acid quantitative test results showed significant decrease (P<0. 05) in 14 d. 3 samples went below the limit of detection.The HCV-RNA readings (IU/m L) at 4/24/48/72 h/7/14 d are as follows: 36. 9; 38. 0, 32. 3, 40. 4, 31. 5; 26. 7, 43. 8, 50. 8, 59. 9, 44. 3; 37. 9, 25. 3, 63. 4, 55. 5, 74. 6; 50. 5, 19. 2, 31. 8, 95. 6, 17. 1; 39. 8, 30. 7, < 15. 0, < 15. 0, <15. 0. Conclusions these data demonstrated that serum samples with HCV-RNA levels lower than 102 IU/m L can be detected in less than 7 days if stored at 4℃. If stored at-20℃, no more than 3 freeze-thaw cycles are allowed to maintain valid test results.
