Abstract
Objective The influence of storage temperature and duration on coagulation the factor Ⅷ activity in fresh plasma was observed in order to propose appropriate frozen procedure to acquire quality fresh frozen plasma. Methods After apheresis, the fresh plasma was immediately divided into 9 aliquots and added into 9 test tubes (20 m L per tube) and stored in the following conditions: room temperature (RT) /0 h, 4℃ 4 h, 4℃ 6 h, 4℃ 8 h, 4℃ 12 h, 4℃ 24 h, RT 4 h, RT 6 h, RT 8 h, RT 12 h, and RT 24 h. The samples were processed to detect FⅧ: C immediately after melting in a 37℃ water bath. Results The coagulant factor VIII activity in fresh plasma decreased with the prolongation of storage time. There was no statistically significant difference in coagulant factor VIII activity between the 0 h storage group and the 4 h or 6 h storage at 4℃/RT (P>0. 05). However, comparing to the value at 0 h, coagulant factor VIII activity decreased significantly when fresh plasma was stored for 8 h, 12 h and 24 h at 4℃/RT (P<0. 05 or 0. 01). The decline rates of coagulant factor VIII activity was lower in the 4℃ stored groups than the RT groups given the same storage period. However, no significant difference (P>0. 05) between the 4℃ and the RT groups were found except for the 24 h storage group (P<0. 01). Conclusion The coagulant factor VIII activity was not affected dramatically if fresh frozen plasma was prepared within 4 h. With the prolonged storage time, the activity showed a decreasing trend. Therefore, we recommend the fresh plasma to be stored at 4 ℃/RT within 6 h to maintain quality blood components.关键词
