一株猪源肺炎克雷伯杆菌的分离与鉴定
Isolation and identification of a strain of Klebsiella pneumoniae from pig

Abstract
A suspected Klebsiella strain was isolated from pork by chance in our laboratory. In order to identify the suspected bacteria,the suspected bacteria were analyzed by means of bacterial isolation and culture,physicochemical properties, 16S rRNA sequence analysis,PCR amplification and bacterial pathogenicity test. The antibiotic sensitivity test results showed that the isolate was highly sensitive to norfloxacin,chloramphenicol and gentamicin,and was resistant to penicillin and cefalexin. In addition,the results of the urea decomposition test and citrate utilization test were positive,and the results of the hydrogen sulfide test and indole test were negative. The 16S rRNA amplification sequence was compared with the nucleotide sequence of Klebsiella pneumoniae published in GenBank. The homology of the isolated strain and Klebsiella acid was from 40. 1% to 47. 2%,and the nucleotide homology of Klebsiella pneumoniae was from 86. 8% to 99. 9% . The isolated strain was confirmed as Klebsiella pneumoniae and named as GZAS2016. PCR amplification of the phoE gene fragment showed a 368 bp band,which further confirmed that the isolate was Klebsiella pneumoniae. In the animal pathogenicity test,the clinical symptoms of SPF mice began to appear at hour 2 after inoculation,and died within 12—15 h. It was proved that the isolated strains had certain pathogenicity to mice. The above-mentioned results showed that a pathogenic Klebsiella pneumoniae strain GZAS2016 was isolated from pork.关键词
