Commensal Lactococcus lactis Accelerates the Development of Drosophila melanogaster

Abstract
To isolate lactic acid bacteria from Drosophila melanogaster gut and investigate their effects on the developmental timing of its hosts,bacteria were isolated from the fly gut with MRS medium and further subjected to 16S rRNA gene sequence identification.The developmental timing and the growth rate of D.melanogaster were assayed.Immunofluorescent staining and real-time quantitative PCR were used to analyze the intestinal cell proliferation and activation of prothoracicotropic hormone (PTTH) and insulin signaling,respectively.Glucose oxidase method was used to determine the concentration of glucose.The results showed that a Lactococcus lactis strain was isolated from D.melanogaster and it efficiently colonized the gut of D.melanogaster.The bacterium facilitated the developmental timing of D.melanogaster by increasing the growth rate.At the cellular level,the number of mitotic cells in L.lactis-associated fly guts was approximately six times as much as that in germ-free (GF) sibling.And at the molecular level,the settled L.lactis activated the hormonal and insulin signals to accelerate the systemic growth of the fruit flies.In conclusion,L.lactis is one of the commensal bacteria of D.melanogaster,and it can stimulate the development of D.melanogaster through hormonal and insulin signals.关键词
