摘要

Abstract

Objective To study the protective effect and its mechanism of ligustilide on ulcerative colitis -induced by trinitrobenzene sulfonic acid (TNBS) in rats.Methods The rats were randomly divided into 5 groups(n=12):blank group, model group, low-, middle-, and high-dose experimental groups ( ligustilide 10, 20, 40 mg· kg -1) .The model of ulcerative colitis was established by injected TNBS 100 mg· kg -1and 50% ethanol into colon of rats except for the blank group, then drugs was given intragastrically for 14 d since the third day after modeling.The index of disease activity was observed .The levels of interleukin-4 ( IL -4 ), interleukin -10 ( IL -10 ) and tumor necrosis factor-α( TNF-α) in serum were determined by enzyme -linked im-munosorbent assay .Results The disease activity indices of the blank group, the model group and the high , medium and low dose groups were (0.33 ±0.14 ), ( 1.83 ±0.49 ), ( 0.94 ±0.27 ), ( 1.18 ±0.33 ), (1.35 ±0.30 ) point.The levels of IL -4 in serum of these 5 groups were (118.27 ±15.23 ), ( 51.35 ±9.80 ), (93.04 ±13.26), (86.31 ±11.92) and (73.09 ±10.57)ng· L-1.The levels of IL-10 in serum of these 5 groups were (85.63 ±10.05 ), (38.13 ±5.86 ), ( 70.92 ±8.87 ), ( 62.09 ±9.28 ) and (53.38 ±7.41 ) ng· L-1.The levels of TNF -α in serum of these 5 groups were ( 43.63 ±5.97 ), ( 81.25 ±11.36 ), ( 52.89 ±6.22 ), (63.30 ±7.83 ), and (72.85 ±10.73 ) ng· L-1, respectively.Compared with the blank group , the differences of all indexes in the model group were statistically significant (all P<0.01).Compared with the model group , all indexes in the high and middle dose experimental groups were statistically significant (P<0.05, P<0.01).Conclusion Ligus-tilide had a protective effect on rat with ulcerative colitis , and its mechanism may be related to up -regulation of IL-4, IL-10 and down-regulation of TNF-α.
