Effects of Dietary Pectic Oligosaccharide on Ileal Barrier Function of Weaned Piglets Challenged by Rotavirus

Abstract

This experiment was conducted to determine the effects of dietary pectic oligosaccharide ( POS) on the ileal barrier function of weaned pigs challenged by rotavirus ( RV ) . Twenty-eight healthy crossbred ( Duroc×Landrace×Yorkshire) piglets weaned at 21 days of age were used in a 2×2 factorial design. The trial factors included POS treatment ( control diet or supplementing 200 mg/kg POS diet) and RV challenge ( oral gavage with RV or sterilized medium) . On day 15, RV challenge was carried out. The trial duration was 18 days. The results showed that RV infection increased the diarrhea rate and positive rate of rotavirus non-struc-tural protein 4 ( NSP4) in ileal mucosa, significantly decreased the villus height and villus height to crypt depth ratio of ileal mucosa ( P<0.05) , significantly reduced the expressions of zonula occludens-1 ( ZO-1) , occlu-din, mucin 1 and mucin 2 in ileal mucosa ( P<0.05) , significantly enhanced the malondialdehyde content in il-eal mucosa and E. coli population in ileal digesta ( P<0.05) , and significantly decreased total antioxidant ca-pacity in ileal mucosa and the population of Lactobacillus and total bacteria in ileal digesta of weaned pigs ( P<0.05) . Dietary POS reduced the diarrhea rate and positive rate of rotavirus NSP4 in ileal mucosa of the RV-in-fected weaned piglets, and significantly alleviated the effects of RV challenge on ileal mucosa morphology, mi-crobial population in ileal digesta, antioxidant capacity and expressions of occludin, mucin 1 and mucin 2 in il-eal mucosa of weaned piglets ( P<0.05) . These results suggest that dietary POS can prevent the RV-induced diarrhea, which is associated with the improvement of ileal barrier function of weaned pigs.
