Effect of Schisandrin B on IL-1β-induced Apoptosis of Nucleus Pulpo-sus Cells by Regulating p38 MAPK Signaling Pathway

Abstract
Objective To investigate the effect and mechanism of schisandrin B ( Sch B) on interleukin-1β (IL-1β) -induced apoptosis of nucleus pulposus ( NP) cells.Methods Primary rat NP cells were cultured and divided into control group, IL-1β group, and Sch B groups (5, 10, 20 μmol/L) .Cells were treated with IL-1β except those in the control group.MTT was used to detect cell proliferation, and Hoechst staining to detect apoptosis.Superoxide dismutase ( SOD ) and malonaldehyde ( MDA) concentrations were measured with the testing kits.Western blotting was performed to detect the levels of Ki67, cleaved caspase-3, Bcl-2, Bax and intervertebral disc degeneration related protein metalloproteinase-13 ( MMP-13 ) , collagen II, integrin-like metallo-proteinase and thrombin type 4 antibody ( a disintegrin and metalloproteinase with thrombospondin motif-4, ADAMTS-4) , proteoglycan and p38 mitogen-activated protein kinase ( p38 MAPK) and p-p38 MAPK expression.Results The cell proliferation rate and the expression levels of Ki67 and Bcl-2 were significantly decreased, and the apoptosis rate and the expression levels of cleaved Caspase-3 and Bax were significantly increased in the IL-1β group when compared with those in the control group.Sch B (5, 10, 20 μmol/L) could significantly reverse the changes of these indexes induced by IL-1β.Meanwhile, the concentration of SOD was significantly decreased and that of MDA was significantly increased in IL-1β group as compared with the control group.The SOD con-centration was significantly higher and the MDA concentration significantly lower in the Sch B (5, 10, 20 μmol/L) group than in the IL-1β group.In addition, IL-1β was found to significantly pro-mote the expression of MMP-13 and ADAMTS-4 in NP cells, increase the ratio of p-P38/P38, in-hibit the expression of collagen II and aggrecan, which was significantly reversed by Sch B (5, 10, 20 μmol/L) .Conclusion Sch B can alleviate IL-1β-induced cell apoptosis of NP cells, and the mechanism may be related to activation of p38 MAPK signaling pathway.关键词
