摘要

Abstract

Objective To explore the effects of astragaloside IV ( AS-IV) on renal fibrosis in rats with diabetic nephropathy.Methods SD rats were divided into a normal control group, model group and AS-IV treatment group (low, middle and high dose groups at 20, 40 and 80 mg/kg re-spectively) .A diabetic rat model was established by using streptozotocin ( STZ) .After 8 weeks of continuous administration, the body mass, kidney index, blood glucose level, serum creatinine ( SCr), urea nitrogen ( BUN) , malondialdehyde ( MDA) , 24-h urinary microalbumin excretion rate (24 h UAER) and glutathione peroxidase ( GSH-Px) and serum total superoxide dismutase ( T-SOD) activity were measured in the rats.The renal histopathological changes were observed in all rats.The protein expression of renal cortical transforming growth factor-β1 ( TGF-β1) , nuclear factor erythroid related factor 2 ( Nrf2) and heme oxygenase 1 ( HO-1) were detected.Results Compared with normal control group, the body mass, kidney index, blood glucose content, glo-merular positive score and 24 h UAER, BUN, MDA amd SCr levels were significantly increased in model group ( P<0.05) while the GSH-Px and T-SOD activity was significantly decreased ( P<0.05) .Moreover, the total protein and nuclear protein expression levels of TGF-β1 and Nrf2 were significantly increased ( P <0.05 ) .Compared with model group, the kidney index, blood glucose level, glomerular positive score and 24 h UAER, BUN, MDA and SCr in middle and high dose groups were significantly decreased ( P<0.05) , and the GSH-Px and T-SOD activity was sig-nificantly increased in high dose group ( P<0.05) , and the expression of TGF-β1 was significant-ly decreased while the expression levels of total protein and nuclear protein of Nrf2 and HO-1 protein were significantly increased ( P<0.05) .Conclusion AS-IV can reduce the liver fibrosis in rats with diabetic nephropathy by increasing serum antioxidant capacity, decreasing TGF-β1 protein ex-pression, activating Nrf2 signaling pathway and enhancing the expression of antioxidant protein HO-1.
