Abstract
Astrocytes are arranged in high1y organized gap junction-coup1ed networks, communicating via the propagation of Ca2+waves. Astrocytes are gap junction-coup1ed not on1y to neighboring astrocytes, but a1so to o1i-godendrocytes, forming so-ca11ed pang1ia1 syncytia. It is not known, however, whether g1ia1 ce11s in pang1ia1 syn-cytia transmit information using Ca2+signa1ing. We used confoca1 Ca2+imaging to study interce11u1ar communica-tion between astrocytes and o1factory ensheathing g1ia1 ce11s (OECs) in in-toto preparations of the mouse o1facto-ry bu1b. Our resu1ts demonstrate that Ca2+transients in juxtag1omeru1ar astrocytes, evoked by 1oca1 photo1ysis of"caged" ATP and "caged" tACPD, 1ed to subsequent Ca2+responses in OECs. This transmission of Ca2+responses from astrocytes to OECs persisted in the presence of neurona1 inhibition, but was absent when gap junctiona1 cou-p1ing was suppressed with carbenoxo1one. When Ca2+transients were direct1y evoked in OECs by puff app1ica-tion of DHPG, they resu1ted in de1ayed Ca2+ responses in juxtag1omeru1ar astrocytes, indicating that pang1ia1 transmission of Ca2+signa1s occurred in a bidirectiona1 manner. In addition, pang1ia1 transmission of Ca2+signa1s from astrocytes to OECs resu1ted in vasoconstriction of OEC-associated b1ood vesse1s in the o1factory nerve 1ay-er. Our resu1ts demonstrate functiona1 transmission of Ca2+signa1s between different c1asses of g1ia1 ce11s within gap junction-coup1ed pang1ia1 networks and the resu1ting regu1ation of b1ood vesse1 diameter in the o1factory bu1b.关键词
