Abstract
Objective To observe the effects of cigarette smoke extract on the mineralization ability of human jawbone-derived osteoblasts in vitro. Methods Osteoblasts were obtained from 30 healthy subjects with dental implants in the Department of Stomatology, Beijing Stomatological Hospital and the General Hospital of Civil Aviation, Affiliated to Capital Medical University, and then and isolated. The final concentration of extract was 0. 5% (low concentration group) and 3% (high concentration CSE group), and smoke-free extract was used as negative control group. The proliferation ability was detected by MTT assay, the in vitro mineralization ability of human jawbone-derived osteoblasts was detected by alkaline phosphatase staining method, and the expression of ALP, BSP and COL-1 in each group was detected by RT-PCR. Results There was no significant difference in the proliferation and mineralization of jawbonederived osteoblasts between control group and low-concentration CSE group (P> 0. 05), and there was also no significant difference in the expression of ALP, COL-1 and BSP (P> 0. 05). The proliferation and mineralization ability of osteoblasts in high concentration group were lower than those in control group and low concentration CSE group (P < 0. 05), and the expression of ALP, COL-1 and BSP was also lower than that of control group and low concentration group (P < 0. 05). Conclusion CSE can affect the formation of osteoblast mineral scaffolds and the deposition of minerals, reduce the quantity and quality of new bone formation, and reduce the degree of calcification of extracellular matrix of osteoblasts, thus affecting the osseointegration of implants.关键词
