摘要

Abstract

A snail enzyme-assisted extraction of polysaccharides from Baiyaqilan tea was used to improve the utilization ratio of Baiyaqilan tea. Based on the single-factor experiment, a four-factor and three-level experiment was designed by the Box-Behnken method using extraction yield of polysaccharides as response value, enzymolysis temperature, enzyme dosage, enzymolysis time and pH as factors. The enzymatic extraction technology of polysaccharide from Baiyaqilan tea was optimized using response surface methodology by Design-Expert 8.05 b software. The scavenging capacity of DPPH, OH and O2- free radicals was used to evaluate its antioxidant activity. The results showed that the optimum extraction parameters were as follows: temperature 51℃, enzyme dosage 1.2%, enzymolysis time 55 min and pH 5.6, and the extraction yield was up to 93.81 mg·g-1. The extraction rate of this process increased 77.74 % than that of the hot water extraction without snail enzyme. Experiment results showed that there was a significant dose-effect relationship between the scavenging effects of polysaccharides from DPPH, OH and O2- free radicals, the IC50 values of DPPH, OH and O2- free radicals were 80.65, 204.39 and 428.10 mg·L-1, respectively.
