Abstract

Objective To optimize the high-efficiency, high-pressure differential and low temperature continuous extraction, isolation and concentration technology and complete equipment to extract polysaccharides from Dendrobium candidum in pilot test and the medium test, in order to provide a reference for large-scale production. Methods With the extraction rate of polysaccharides from Dendrobium candidum as the evaluation index, the orthogonal test L9 (34) was used to investigate the three factors which influenced the extraction process of polysaccharides from Dendrobium candidum, such as grinding degree, ratio of dried powder of Dendrobium candidum to water and extraction pressure. Results The optimal parameters of extraction process were as follows: grinding degree of 1.2 mm, 1:25 (g:mL) ratio of dried powder of Dendrobium candidum to water, extraction pressure of 40 MPa. Under the conditions, the extraction rate of polysaccharides was 48.5%, and the extraction rate of medium test was 45.0%. Conclusion The extraction technology optimized by orthogonal test is stable, simple and high extraction rate, which has a certain guiding significance for the large-scale production of polysaccharides from Dendrobium candidum.
