Abstract
Objective To compare the difference of HPLC characteristic spectra of Phragmitis Rhizoma extract and Zizaniae caduciflorae Rhizoma extract, so as to identify the Phragmitis Rhizoma extract and its adulterants. Methods HPLC was performed on ZORBAX SB C18 column (250 mm×4.6 mm, 5 μm), the acetonitrile-0.5% potassium dihydrogen phosphate aqueous solution (pH2.15) was employed as a mobile phase at gradient elution, detection wavelength was 275 nm;column temperature was 30℃, and flow rate was 1.0 mL/min. The contrast spectrum was generated with Traditional Chinese medicine (TCM) chromatographic fingerprint similarity evaluation software system (2004 A version) by State Pharmacopoeia Commission. Results Seven common peaks were separated from 10 batches of Phragmitis Rhizoma extract, including p-coumaric acid and ferulic acid. Thirteen common peaks were separated from 10 batches of Zizaniae caduciflorae Rhizoma extract, which included 7 same common peaks of Phragmitis Rhizoma extract and six common peaks (peak w1-peak w6) of flavonoids easily recognized. The similarities of 10 batches of Phragmitis Rhizoma extract and Zizaniae caduciflorae Rhizoma extract were 0.935-0.999, 0.807-0.973 respectively, and the similarity of the mutual modes between Phragmitis Rhizoma extract and Zizaniae caduciflorae Rhizoma extract was 0.875. Conclusion The method is accurate, reliable and reproducible, which can be used for the identification and quality evaluation of Phragmitis Rhizoma extract.关键词
