葡聚糖硫酸钠诱导慢性溃疡性结肠炎小鼠模型的建立及评价
Establishment and evaluation of chronic ulcerative colitis mice model induced by dextran sodium sulfate

Abstract
Objective:To establish and evaluate chronic ulcerative colitis(UC) mice model induced by different concentrations of dextran sulfate sodium (DSS).Methods:Female C57BL/6mice were randomly divided into normal control group,2.0％ DSS group,2.5％ DSS group,and 3.0％ DSS group.The mice in normal control group were given orally distilled water.Mice in 2.0％ DSS group,2.5％ DSS group,and 3.0％ DSS group were respectively given 2.0％ DSS,2.5％ DSS and 3.0％ DSS via gavage on day 1-5,11-15and 21-25. The mental status,mortality,body weight and the disease activity index(DAI) were detected.All mice were sacrificed on day 30,and colon tissues were collected for HE staining.Results:Except for the normal control group,other groups showed obvious clinical symptoms and pathological changes.The total histological scores in 2.0％ DSS group,2.5％ DSS group,and 3.0％ DSS group were higher than that in the normal control group(P<0.01).Compared with the normal control group,the DAI score of 2.0％ DSS group, 2.5％ DSS group and 3.0％ DSS group was increased on the 2nd day,and reached the highest on the 6th day,then decreased gradually.It elevated again on the 10th day,began to decline on the 14th day,approached the normal level on the 20th day,rose again after the 20th day and notably decline on the 24th day(P<0.05).Conclusion:The chronic UC mice model was successfully induced by 2.0％ DSS,2.5％ DSS and 3.0％ DSS.The application of 2.5％ DSS resulted in the lowest mortality and the best stability.关键词
