Abstract

To explore the effect of Antithrombosis and Channel-Unblocking Pills on myocardium of rats with ischemia-reperfusion injury.Methods:Forty healthy male rats were randomly divided into the model group, the diltiazem group, the high-dose TCM group and the low-dose TCM group, with 10 rats in each group.The model group was given 10 mL·kg-1 of saline by intragastric administration, while the high-dose TCM group, the low-dose TCM group and the diltiazem group were given KSTLP with 0.045 g·L-1, 0.180 g·L-1 and 5 mg·kg-1 concentration, and diltiazem of 5 mg·kg-1 plus sodium chloride solution respectively.Myocardial ischemia-reperfusion injury model in rats was established by ligating the anterior descending branch of left coronary artery after one week of intragastric administration.The changes of heart rate and ST segment during ischemia-reperfusion injury in each group were monitored by dynamic lead II electrocardiogram, and the levels of interleukin-6 (IL-6) in serum were measured by enzyme-linked immunosorbent assay (ELISA).Results:Compared with the model group, the high-dose TCM group and the diltiazem group could effectively slow down the heart rate in different time periods of ischemia-reperfusion injury (P<0.05).There was no significant difference between the low-dose TCM group and the model group (P>0.05).Compared with the model group, the decrease of ST segment in the high-dose TCM group and the diltiazem group was the most obvious.There was significant difference in different time periods of ischemia from the 20th minute (P<0.05), while there was no significant difference in ST segment between the low-dose TCM group and the model group at different time points of ischemia-reperfusion injury (P>0.05).Compared with the model group, the high-dose TCM group, the low-dose TCM group and the diltiazem group could reduce the level of IL-6 in serum caused by ischemia-reperfusion injury.The high-dose TCM group was higher than the low-dose TCMgroup, and the difference was statistically significant (P<0.05).Conclusion:Antithrombosis and Channel-Unblocking Pills can improve myocardial ultrastructure and abnormal electrocardiogram in ischemia-reperfusion injury, reduce the content of IL-6, inhibit inflammation, and play a protective role in myocardium.
