不同离心力分离外周血单个核细胞的条件优化
Condition optimization of different centrifugal force on separating peripheral blood mononuclear cells

Abstract
Objective To observe the effect of different centrifugal force on the separation of peripheral blood mononuclear cell (PBMC) .Methods PBMCs were isolated by adopting the ficoll density gradient centrifugation at different centrifugal force of separation and centrifugal force of washing.Then the yield rate and survival status of PBMCs were measured by flow cytometry and the statistical analysis was performed.Results The results of separation centrifugal force:the maximum number of PBMCs was obtained at 600 g.The number of apoptotic cells between 300-900 g separation in detection at 24 hhad no statistically significant difference, moreover was relatively less, increasing centrifugal force, the number of apoptosis cells was increased;in the detection at 72 h, the centrifugal force higher than 1 200 g or low to 300 g, the survival cells were significantly decreased.The results of washing centrifugal force:the centrifugal force was 150 g, the yield rate of PBMCs was significantly lower than that of 300-2 400 g.The apoptotic cells detected at 24 hshowed that the centrifugal force was more than 600 g, the number of apoptosis cells was increased;the detection at 72 h showed that the washing centrifugal force was more than 1 200 g, and the number of survival cells was decreased significantly.Conclusion The centrifugal force of separation is 600-900 g and centrifugal force of washing is 300-600 g, which all can be used for the separation of PBMC.关键词
