Abstract

Objective To investigate the effect of miR-98 on the biological behavior of nasopharyngeal carcinoma (NPC) cells and its mechanism. Methods Twenty-five patients with NPC who were treated in our hospital were enrolled. Cancerous tissues and adjacent normal tissues 1 cm away from the tumor were collected from each patient and NPC cell line CNE1 used for subsequent experiments was divided into control group (NC group) and experimental group (miR-98-inhibitor group). The NC group was transfected with NC lentivirus mimics and miR-98-inhibitor group was transfected with miR-98-inhibitor mimics. Tumor xenograft models were prepared from 4-week-old female nude mice. The expressions of miR-98 in NPC tissues, adjacent normal tissues and different NPC cell lines (CNE1, TW03 and NP69) were detected. The relationship between miR-98 and BACH1 was analyzed, and the influence of miR-98 on invasion ability of NPC cells were observed. The effect of miR-98 D on the tumorigenicity of NPC cells in nude mice was recorded. Results The expressions of miR-98 in adjacent normal tissues and NPC tissues were 2.05±0.28 and 3.98±0.56 respectively, and there were significant differences (P<0.05). The expressions of miR-98 in NPC cell lines CNE1, TW03 and NP69 were 5.21±0.43, 2.29±0.21 and 0.56±0.10, respectively, and significant differences were found (P<0.05). miR-98 significantly inhibited the activity of BACH1-3'UTR wild-type dual luciferase. The number of invasive cells in NC group and miR-98-inhibitor group was 191.36±23.98 and 41.09±8.75, respectively. The tumor volume of nude mice was (4.32±0.34) cm3 and (1.02±0.09) cm3, and the weight was (4.16±0.29) g and (0.96±0.06) g, respectively, suggesting significant differences (P<0.05). The positive expression rate of BACH1 protein in cancerous tissues of nude mice in miR-98-inhibitor group was significantly lower than that in NC group (P<0.05). Conclusion miR-98 can regulate the expression of BACH1, thereby affecting the biological behavior of NPC cells.
