人良性乳腺病血清双向电泳图谱的建立和分析
Establishment and analysis of two-dimensional electrophoresis maps for serum samples of patients with benign breast disease

Abstract
Objective: To identify the serum proteins differentially expressed between patients with benign breast disease (BBD) and healthy controls through two-dimensional electrophoresis (2-DE) and mass spectrometry, and to preliminarily explore the tumor markers for early diagnose of BBD. Methods: The serum samples of 10 BBD patients and 10 healthy controls were collected. Then the experimental conditions of 2-DE were established, and serum proteins were separated under the same conditions. Protein spots expressed differentially between the two groups were screened out by PDQuest software and identified by mass spectrometry. Results: The 2-DE maps of BBD patients and healthy controls were preliminarily established, and 12 protein spots differentially expressed between the two groups were detected by PDQuest analysis of the 2-DE data. Finally, one differentially expressed protein spot was identified successfully by mass spectrometry. The spot corresponded to two proteins, i.e., Rho-associated protein kinase 1 and keratin-10. Conclusion: Rho-associated protein kinase 1 and keratin-10 may be the tumor markers for early diagnosis of BBD.关键词
