Correlation between SHC-mediated insulin signal transduction MAPK pathway and congenital heart disease

Abstract
Objective To observe the correlation between the abnormal insulin signal transduction pathway and the occurrence of no syndrome-congenital heart disease (ns-CHD). Methods The right atrium tissues from 5 cases of nsCHD patients (case group) and 2 cases of rheumatic heart disease (RHD) patients with mitral stenosis (MS) were detected by digital gene expression profiling (DGE), then we verified them in the right atrium tissues from 37 cases of ns-CHD patients and 12 cases of RHD MS patients by using real-time fluorescence quantitative PCR (RT-QPCR). Results SHC gene family-mediated MAPK pathway was restrained in the case group, and the expression of 6 genes (PTPRF, SHC4, ARAF, MAP2 K2, MKNK2, and ELK1) was significantly down-regulated (all P < 0. 05). Conclusions Hence SHC-mediated MAPK pathway may influence the heart development. The relationship between SHC and insulin resistance may be one of the reasons for the high rate of neonatal CHD in gestational diabetes mellitus (GDM) neonates.关键词
