摘要

Abstract

Objective To initially explore the up-regulated expression mechanism of repressor element 1 silencing transcription factor (REST) in the intermediate-conductance Ca2 +-activated K+channel (KCa3. 1) of the cervical squamous cell carcinoma, and to provide a new site for the study of the occurrence and development of cervical squamous cell carcinoma. Methods The cervical squamous cell carcinoma tissues, paracancerous tissues, and normal cervical tissues were collected, and REST protein and mRNA expression in different tissues was detected by immunohistochemistry and qRT-PCR. ChI P-qPCR was used to detect the enrichment degree of REST at the site of repressor element l (RE1) of KCa3. 1 promoter region. Results The relative expression levels of REST gene in the cervical squamous cell carcinoma, paracancerous tissues and normal cervical tissues were 0. 46 ± 0. 06, 0. 89 ± 0. 13, and 1. 00 ± 0. 12, respectively; compared with the paracancerous tissues and normal cervical tissues, the relative expression of REST gene in the cervical squamous cell carcinoma dropped by 48% and 54%, respectively; the expression of KCa3. 1 gene in the cervical squamous cell carcinoma, paracancerous tissues and normal cervical tissues were 2. 14 ± 0. 35, 1. 18 ± 0. 21, and 1. 00 ± 0. 13, respectively, and the expression of KCa3. 1 gene in cervical squamous cell carcinoma was 81% and 114% higher than that in the paracancerous tissues and normal cervical tissues, respectively. REST protein expression level and mRNA transcription level in the cervical squamous cell carcinoma were significantly lower than those in the normal cervical tissues and paracancerous tissues (both P < 0. 05). The expression of KCa3. 1 in the cervical squamous cell carcinoma was significantly higher than that in the paracancerous tissues and normal cervical tissues. REST could bind to RE1 site of KCa3. 1 promoter region, and the the enrichment degree of REST at RE1 site of KCa3. 1 promoter region in the cervical squamous cell carcinoma tissues was lower than that in the normal cervical tissues and paracancerous tissues (P < 0. 05). Conclusions REST negatively regulates KCa3. 1 expression by binding to RE1 site of KCa3. 1 promoter region. The down-regulation of REST expression decreases the inhibition of KCa3. 1 gene expression, increases the expression of KCa3. 1 gene, enhances the activity of KCa3. 1, and promotes the pathogenesis and development of cervical squamous cell carcinoma.
