Abstract
Objective To investigate the expression changes of glucose transporter-1 (GLUT-1) and pyruvate kinase M2 (PKM2) in the gastric carcinoma BGC-823 cells after we used the small interfering RNA to silence hypoxia inducible factor-1α (HIF-1α) under hypoxia. Methods BGC-823 cells were divided into four groups: the normal group, hypoxic control group, HIF-1α siRNA-transfected group (HIF-1α siRNA group) and control siRNA-transfected group (control siRNA group). Cobalt chloride (Co Cl2) was used to simulate the hypoxic environment of tumor cells in vitro. The hypoxic control group was cultured with 200 μmol/L Co Cl2 for 12 h. The HIF-1α siRNA group and the control siRNA group were transfected with HIF-1α-siRNA and Control-siRNA for 48 h and then were cultured with 200 μmol/L Co Cl2 for 12 h. The expression levels of HIF-1α, GLUT-1, and PKM2 mRNA and protein in the cells of the four groups were detected by quantitative real-time PCR (qRT-PCR) and Western blotting. Results Compared with the normal group, the expression of HIF-1α mRNA in the hypoxia control group did not change significantly (P> 0.05) , while the expression of HIF-1α protein increased, and the mRNA and protein expression of GLUT-1 and PKM2 increased (P < 0.05 or P < 0.01). Compared with the hypoxic control group, the mRNA and protein expression of HIF-1α, GLUT-1, adn PKM2 in the HIF-1α siRNA group decreased (P < 0.05 or P < 0.01) , and there was no significant change in the mRNA and protein expression of HIF-1α, GLUT-1 and PKM2 in the control siRNA group (all P> 0.05). Compared with the control siRNA group, the mRNA and protein expression of HIF-1α, GLUT-1, and PKM2 in the HIF-1α siRNA group all decreased (P < 0.05 or P< 0.01). Conclusion Hypoxia can promote the expression of HIF-1α, GLUT-1 and PKM2 in the human gastric cancer BGC823 cells, the expression of HIF-1α, GLUT-1, and PKM2 is down-regulated after silencing HIF-1α, and HIF-1α maybe involved in regulating the expression of GLUT-1 and PKM2.关键词
