PMA-qPCR快速检测畜禽肉类中沙门氏菌活菌方法的建立
Establishment of PMA-qPCR assay for rapid detection of viable Salmonellain livestock and poultry meat

Abstract
A Propidium Monoazide-quantitive Polymerase Chain Reaction (PMA-qPCR) method was established to quantitatively detect live Salmonella in livestock and poultry meat.The light exposure time and the concentration of PMA were optimized.The standard curve was established by standard plasmid.The sensitivity and specificity of this assay were investigated respectively.The results showed that the amplification of DNA derived from the dead Salmonellacells could be inhibited without affecting the viable cells when PMA was at a dose of12μg/mL and the light exposure time of 6min.In artificial pork samples, PMA-qPCR could detect as low as10CFU/mL.It was possible to rapidly and quantitatively detect viable Salmonella enteritidis in meat of livestock and poultry by the PMA-qPCR assay.关键词
