阿替普酶静脉溶栓治疗急性脑梗死大鼠的疗效及对脑组织炎性因子表达的影响

Abstract
Objective To explore the effect of antiprase (r-tPA) intravenous thrombolysis in the treatment of acute cerebral infarction rats and the influence on the expression of inflammatory factors in brain tissue. Methods Forty-two acute cerebral infarction rats were randomly assigned to model group (n = 21) and test group (n = 21) ,while 20 healthy rats were assigned to sham-operation group. The test group was treated with r-tPA (5 mg·kg-1) by caudal vein injection within 3 h after infarction,and model group and sham-operation group were treated with equal amount of 0. 9% NaCl. The volume of cerebral infarction after infarction were detected by chloride three phenyltetrazole (TTC) staining,the expression of inflammatory factors in brain tissue were detected by enzyme-linked immunosorbent assay (ELISA). Results On Day 7 after infarction,the volume of cerebral infarction in the model group were (86. 94 ± 4. 08) % and (42. 12 ± 2. 89) %,and the differences was significant (P < 0. 05). On Day 7 after infarction,there were significant diffe-rences among the sham-operation group,model group and the test group in tumor necrosis factor-α(TNF-α) [(0. 32 ± 0. 07) pg·mL-1 vs(1. 36 ± 0. 12) pg·mL-1 vs(0. 27 ±0. 10) pg·mL-1] and intercellular adhesion molecule-1 (ICAM-1) [(1. 39 ± 0. 11) pg ·mL-1 vs(28. 79 ±2. 12) pg·mL-1 vs (17. 32 ±1. 91) pg·mL-1](all P <0. 05). Conclusion r-tPA intravenous thrombolysis treatment can effectively reduce the volume of cerebral infarction and the expression of TNF-α,ICAM -1 in brain tissue of acute cerebral infarction rats.关键词
