摘要

Abstract

Objective To investigate the effect of recombinant human granulocyte colony-stimulating factor (rhG-CSF) on angiogenesis and apoptosis after acute ischemic cerebral infarction in rats. Methods Out of 80 rats,55 rats were randomly chosen and assigned to model group and test group [middle cerebral artery embolism (MCAO) by Longa's method]. The other 25 rats were assigned to sham-operation group(underwent same procedure without tethering). The rats in test group were injected subcutaneously with 60 μg·kg-1 rhG-CSF,while both sham-operation group and model group were injected with the same amount of 0. 9% NaCl once daily for 3 days. After cerebral ischemia and reperfusion,the volume of cerebral infarction was measured by 2,3,5-Triphenyl-2H-tetrazolium chloride (TTC) staining,the expression of Survivin and Egr-1 protein were detected by immunohistochemistry,and the apoptosis of neurons was detected by TUNEL. Results After 7 days of cerebral ischemia and reperfusion,of sham-operation group,model group and test group,the infarct volumes were 0%,(30. 75 ± 3. 53) %,(22. 59 ± 3. 42) %,respectively; Survivin protein immunohistochemical scores were 0. 54 ± 0. 23,1. 48 ± 0. 36,2. 41 ± 0. 46,respectively; Egr-1 protein immunohistochemical scores were 0. 84 ± 0. 25,1. 78 ± 0. 52,2. 62 ± 0. 33,respectively; the apoptotic rates of neurons were (1. 36 ± 0. 27) %,(50. 77 ± 2. 48) %,(30. 64 ± 1. 32) %,respectively. There were significant differences between the test group and the model group (all P < 0. 01). Conclusion rhG-CSF can reduce the cerebral infarct volume and neuronal apoptosis after acute ischemic cerebral infarction in SD rats,and up-regulate the expression of Survivin and Egr-1 protein in brain tissue to promote angiogenesis,and it has significant neuroprotective effect.
