Abstract
Objective To evaluate the effect of acute blood loss on the NSE and Claudin-5 content in the serum of rats with mechanical ventilation. Methods Thirty Wistar rats, weighing 300-350 g, were randomly divided into 5 groups according to the volume of blood loss (n=6): group C, control group: no blood loss; group S2, 20% of blood loss; group S4, 40% of blood loss; group S6, 60% of blood loss; group S8, 80% of blood loss. After anesthesia, bloodletting was administered in each rat according to its corresponding groups from its carotid artery; fluid resuscitation was performed from the femoral vein in the meantime. 3 h 55 min after resuscitation, blood samples were collected immediately for artery blood gas analysis. Blood oxygen content was calculated according to oxygen partial pressure and hemoglobin content. After bleeding for 24 hours, 1 mL of blood were taken from carotid artery in each group for examining the contents of NSE and Claudin-5 in the serum. Results 1) Compared with group C, the contents of blood oxygen of the other groups were decreased (P<0.05). Group S8 was the lowest group (6.3±0.4) mL/dl. And the contents gradually decreased as the amount of blood loss increased: S8<S6<S4<S2) The differences were statistically significant (P<0.05). 2) The contents of NSE in the S2, S4, S6 and S8 group were higher than that in group C (P<0.05). Group S8 was the highest one (19.8±0.4) ng/mL with the following order: S8>S6>S4>S2, (P<0.05). The levels of Claudin-5 in group S4, S6 and S8 were lower than that in group C (P<0.05) with group S8 being the lowest one (0.4±0.1) ng/mL and S8<S6<S4, (P<0.05). Conclusion Serum NSE increased after 20% of blood loss for 24 h; serum Claudin-5 decreased after 40% of blood loss for 24 h. Rats cannot survive an 80% of blood loss for 24 h.关键词
