Expression of SLC5A8 Gene in Primary Hepatocellular Carcinoma and Its Relationship with the Prognosis

Abstract
Objective To investigate the expression of SLC5 A8 protein in hepatocellular carcinoma(HCC) and its relationship with the prognosis of HCC. Methods 97 cases of paraffin-embedded hepatocellular carcinoma (HCC) were selected. The expression of SLC5A8 protein was detected by immunohistochemical Elivision plus method. The expression of SLC5A8 under different factors was analyzed. Combined with the postoperative follow-up data, the relationship between SLC5A8 gene and prognosis of HCC was studied. Results The expression of SLC5A8 protein in HCC tissues was significantly lower than that in paracancerous tissues and normal liver tissues (P＜0.01). The expression of SLC5A8 protein in HCC tissues with tumor diameter ≤5 cm, well / moderate differentiation, no cirrhosis, AFP ≤400 ng / ml, tumor grade Ⅰ～Ⅱ was higher than that of tumor diameter ＞5 cm, poor differentiation, liver cirrhosis, AFP ＞400 ng / ml, and tumor grade Ⅲ～Ⅳ (P＜ 0.05). There was no significant difference in the expression of SLC5A8 protein in primary hepatocellular carcinoma(HCC) with different ages, genders and HBsAg (negative or positive) (P＞ 0.05). Kaplan-Meier survival curve analysis showed that the prognosis of patients with high expression of SLC5A8 was better than that of patients with low expression of SLC5A8, and there was significant difference in median survival time between the two groups (P＜0.001). Conclusion The decrease or deletion of SLC5A8 gene expression plays a certain role in the occurrence and development of HCC, while the liver cirrhosis may be an important related factor in the occurrence of HCC. The patients with high expression of SLC5A8 protein have a relatively long postoperative survival time, which may plays a important role in judging the prognosis of patients with primary HCC after operation.关键词
