Abstract

Objective:To observe the thrombolytic effects and hemorrhage of urokinase (UK), alteplase (rt-PA) and reteplase (rPA) on deep venous thrombosis (DVT) in rabbits.Methods:Rabbit models of deep vein thrombosis were prepared by electrical stimulation and stenosis, and uk, rt-PA and r-PA were administered intravenously.The changes of thrombus weight and bleeding after administration were observed.Activated partial thromboplastin time (APTT), thrombin time (TT), fibrinogen (FIB) were determined using plasma coagulation method, and plasminogen (PLG) levels were measured using chromogenic substrate method.Results:The three thrombolytic agents had significant thrombolytic effects on the thrombus of inferior vena cava in rabbits, and the ability of thrombolytic were r-PA>rt-PA>UK.The three thrombolytic agents caused different degrees of bleeding, and the degrees of bleeding were r-PA>rt-PA>UK.Alteplase significantly prolonged TT, reteplase significantly prolonged PT and TT.r-PA and rt-PA significantly reduced the content of FIB in the plasma, and UK and r-PA significantly reduced the content of PLG in the plasma.Conclusion:The three thrombolytic agents had inhibitory effects on deep venous thrombosis, but bleeding also increased significantly with the increase of efficacy.When thrombolytic agents were used for deep vein thrombolysis treatment, bleeding should be pay close attention.
