Effect of Electro-Acupuncture on Neuronal Autophagy in CIRI Rats

Abstract
Objective: To investigate the effects of electro-acupuncture on the expression of autophagy-associated protein LC3A/B in the ischemic penumbra of rats with cerebral ischemia-reperfusion injury (CIRI).Methods: 75 Sprague-Dawley rats were randomly divided into the sham operation group, the model group, the electro-acupuncutre group (EA group), the 3-MA (autophagy inhibitor) group, and EA + 3-MA group;each group was divided into three subgroups of IR24 h, IR72h and IR7d (n = 5). The composite neurological function score, the hanging line test score and the balanced walking test score were measured respectively at the three time points. The expression of autophagy-related protein LC3A/B was detected by immunohistochemistry. Results: In terms of nerve function score, the signs of neurological deficits of MCAO/R rats were alleviated in the EA group; of which there were stastistical differences between the EA group and the model group (P <0. 01); 3-MA group aggravated the signs of neurological deficits when compared to the model group (P <0. 01); however, 3-MA + EA group did not significantly improve the neurological deficit when compared to the model group (P> 0. 05). In terms of the LC3A/B protein expression, it showed significant increase at various time points in the model group compared to the sham operation group (P < 0. 05); it reached the peak at IR72h; the expression of LC3A/B protein in the EA group was significantly higher than that in the model group at IR72h (P < 0. 05); at the time of IR7 d, the expression of LC3A/B protein was significantly lower in the EA group than that in the model group (P < 0. 05); the 3-MA group can significantly down-regulate the expression of LC3A/B protein; electro acupuncture intervention on the basis of the inhibitor, could up-regulate the expression of protein on IR72 h and IR7 d, there were statistical difference between the EA group and the 3-MA group (P < 0. 05). Conclusion: Scalp point-to-point needling combined with electro-acupuncture can significantly improve the neurological deficit of MCAO/R rats, which has neuroprotective effects. It can regulate the expression of LC3A/B protein in the ischemic penumbra of MCAO/R rats, thereby regulating the autophagy of nerve cells. It can increase the expression of LC3A/B protein in the ischemic penumbra in the early stage of cerebral ischemia-reperfusion, moderately activate the autophagy of nerve cells in MCAO/R rats, and protect the damaged neurons; in the mid-late reperfusion period, the expression of LC3A/B protein in the ischemic penumbra was decreased, and the excessive activation of autophagy in the neurons of MCAO/R rats was inhibited, thereby antagonizing further damage of neurons.关键词
