Protection of penehyelidine hydrochloride post-conditioning against damage of rat small intestines mucosal pathological damage caused by limb ischemia reperfusion injury

Abstract

Objective To observe the effect of penehyclidine hydrochloride post-conditioning on the intestinal mucosal barriers in lower limbs of ischemia- reperfusion rats, and to explore the protective mechanism of it against intestinal mucosal damage caused by ischemia reperfusion of limbs. Methods Wistar male rats were randomly divided into 3 groups (36 rats in each group): the sham operation group (group S), the lower limb ischemia reperfusion group (group LIR) and the penehyclidine hydrochloride post-conditioning group (group PHC). The model of lower limb ischemia reperfusion was prepared by the method of using rubber bands to ligate the roots of the rats' hind limbs for 3 h. After complete blocking of blood flow in the lower limbs of rats in the PHC group for 3 h, the rats were reperfused after having been injected with penehyclidine hydrochloride (0.15 mg/ kg) into their tail vein. In immediate (t1), 1 h (t2), 3 h (t3), 6 h (t4), 12 h (t5) and 24 h (t6), wet/dry weight ratios (W/D) and concentrations of malondialdehyde (MDA) were measured after reperfusion. The pathological changes of small intestinal mucosae were observed under an optical microscope, and the pathological grade of Chiu's was also observed. Results Compared with group S, Chiu's grade was increased at each time point in group LIR and PHC (P < 0.05). Compared with group LIR, the pathological grade of small intestinal mucosal Chiu's decreased in group PHC at t3—t6 (P < 0.05). Compared with group S, W/D and MDA content increased at each time point in small intestine of group LIR and PHC (P < 0.05). Compared with group LIR, W/D and MDA content decreased in small intestines of rats in group PHC at t2—t6 (P < 0.05). Conclusion Penehyclidine hydrochloride post-treatment can effectively reduce the damage of ischemia reperfusion on intestinal mucosae, and its mechanism may be related to the inhibition of MDA toxicity on intestinal mucosal cells by penehyclidine hydrochloride.
