长爪沙鼠全脑缺血再灌注损伤后半胱氨酰白三烯受体表达变化的研究
Expression of cysteinyl leukotrienes receptors in gerbils brain after global cerebral ischemia / reperfusion injury

Abstract
Objective To investigate the expression of cysteinyl leukotrienes receptors in gerbils brain after global cerebral ischemia/reperfusion (CI/R) injury. Methods The model of CI/R was established in 110 gerbils by bilateral common carotid occlusion for 10 min followed by 3, 6, 12, 24h- and 3, 7, 14d- reperfusion. After 24h- or 14d- reperfusion, neurological deficit scores were evaluated and the behavioral dysfunction was assessed. The expression of cysteinyl leukotrienes receptors (CysLT1R and CysLT2R) in the homogenate of cerebral cortex and hippocampus were determined by Western blotting. The CysLT1R and CysLT2R positive cells in cerebral cortex and hippocampus were observed by immunohistochemistry. Results Compared to the sham group, the neurological deficit scores were significantly increased and the behavioral dysfunction was aggravated in CI/R gerbils after 24h- or 14d- reperfusion. The expressions of CysLT1R and CysLT2R in gerbil brain were up- regulated after CI/R injury. CysLT1R expression had two peaks at 12h and 14d, respectively, while CysLT2R expression had only one peak at 12h. The CysLT1R and CysLT2R positive cells in cerebral cortex and hippocampus were increased markedly at 24h and 14d. Conclusion CI/R injury induces up- regulation of CysLT1R and CysLT2R in gerbils cerebral cortex and hippocampus, the expression levels were dynamically alterated during 3h- 14d of reperfusion. These findings suggest that CysLTR may be involved in neuron damage and gliosis (glial scar formation) following global cerebral ischemic injury.关键词
