Protective effect of abatacept in on kidney function in rats with type 2 diabetes melitus

摘要

Abstract

Objective To investigate the effect of abatacept on kidney function in rats with type 2 diabetes melittus (T2DM). Methods Forty five male Sprague Dawley (SD) rats were divided into control group, abatacept group and non- intervention group with 15 rats in each group. Type 2 diabetic model was established with high- sugar and high- fat feeding combined with small dose of streptozotocin (STZ) in rats of abatacept group and non- intervention group. The rats in abatacept group were given 0.5mg kg- 1 w- 1 abatacept for 8- 10 weeks. The blood flow parameters of renal main artery determined with color Doppler ultrasound; the elasticity of renal parenchymal was determined with ultrasonic elastography; the blood biochemical indexes were measured, the histology of renal parenchymal was examined with HE staining; the ultrastructure of podocytes was examined with electron microscopy and the expression of B7- 1 in renal tissues was detected with Western blot. Results There were significant differences in peak systolic velocity (PSV), end diastolic velocity (EDV) and mean velocity (MV) among three groups, and these indicators were ranked as nonintervention group <abatacept group <control group (P<0.05). There were significant differences in systolic acceleration (SAC), pulsatility index (PI) and resistance index (RI) among three groups, which ranked as nonintervention group>abatacept group>control group (P<0.05). There were significant differences in the stiffness of kidney among three groups, which ranked as nonintervention group>abatacept group>control group (P<0.05). Compared to the control group, fasting blood glucose (FBG), creatinine clearance rate (Ccr) and urine albumin excretion rate (UAER) in the non- intervention and abatacept groups were significantly increased (P<0.05); while Ccr and UAER in the abatacept group were significantly lower than those in the nonintervention group (P <0.05). HE staining and transmission electron microscope observation showed that there were significant changes in the renal parenchyma of rats in the nonintervention group and the abatacept group, and the changes of kidney in the nointervention group were worse than those in the abatacept group. There were significant differences in the B7- 1 expression in renal tissues among these three groups, which ranked as control group< abatacept group<nonintervention group (P<0.05). Conclusion Abatacept can significantly inhibit the damage of renal function, the increase of resistance and decrease of velocity of renal blood flow, increase of renal parenchyma stiffness, thickening of glomerular basement membrane, reduction of podocytes in diabetic rats, indicating that abatacept can attenuate the renal injury and protect the renal function in type 2 diabetic rats to some extent.
