Effects of β-elemene Combined with Docetaxel on Proliferation and Apoptosis of HeLa Cells in Human Cervical Cancer ★

Abstract
Objective To observe the effects of β-elemene combined with docetaxel on the proliferation and apoptosis of HeLa cell in human cervical cancer. Methods Cell counting kit CCK8 method was used to observe the inhibitory effects of β-elemene combined with docetaxel on the proliferation of HeLa in cervical cancer. Flow cytometry was used to observe the effects of β-elemene combined with do-cetaxel on the apoptosis of HeLa cells. The inverted fluorescence microscope was applied after Hoechst 33258 staining to observe the ef-fects of β-elemene combined with docetaxel on the morphology HeLa cells. Results The inhibitory rates of cell proliferation in combination therapy group at 12 h, 24 h and 48 h were significantly higher than those in docetaxel group and β-elemene group (P<0.05). The interaction coefficients of combination therapy group at 12 h, 24 h and 48 h were all <1, suggesting that β-elemene combined with docetaxel could syn-ergistically inhibit the proliferation of HeLa cells. The apoptosis rate in combination therapy group at 48 h was significantly higher than that in docetaxel group and β-elemene group (P<0.05). The HeLa cell showed obvious apoptosis in combination therapy group. The expression levels of p-MEK1/MEK2 and p-ERK1/ERK2 protein in the combination group were significantly lower than those in the control group and the single drug group (P<0.05). Conclusions β-elemene combined with docetaxel could promote the apoptosis and inhibit the proliferation of HeLa cells. Combination of the two could enhance the anti-tumor activity. The mechanism may be related to the inhibition of the MAPK signaling pathway.关键词
