Effects of 3D Matrigel Model on the Localization of Integrin β1 of Breast Cancer Cells and the Adhesion Ability of Breast Cancer Cells to the Extra-cellular Matrix

Abstract
Objective: To investigate the the effects of matrigel, a 3D culture model, on the morphology, localization of integrin β1 and adhesion ability of breast cancer cells to laminin, fibronectin and collagen IV; study the different interactions of breast cancer cells and extracellular matrix (ECm) in 2D and 3D culture models; and reveal the effect of tumor microenvi-ronment on tumor cells. Methods: mDA-mB-435S and mDA-mB-231, breast cancer cells, were cultured with 2D and 3D matrigel models. The viability of cells was tested by Calcein Am. The localization of integrin β1 was studied with immunos-taining. And the adhesion ability of breast cancer cells to ECm were measured by adhesion assay. Results: The localization of integrin β1 exhibited significant difference between 2D and 3D cultures. mediated by integrin β1 in 3D models, the adhe-sion ability of breast cancer cells to laminin increased significantly and that to fibronectin and collagen IV decreased signifi- cantly. Conclusion: 3D culture model can mimic the 3D microenvironment in vivo and can be used as a new study model to reveal the interaction of tumor cells and ECm and the effects of tumor microenvironment on tumor progression.关键词
