紫苏PfDGAT1酵母功能互补分析
Functional complementation analysis of H1246 yeast mutation by P f DGAT1gene from Perilla f rutes-cens

Abstract
[Objective]In the previous study ,the diacylglycerolacyltransferase (DGA T ) gene P f DGA T1 has been cloned from perilla seeds and highly expressed .In this study ,yeast Saccharomyces cerev isiae mutant strain H1246 was used to test the function of DGA T for catalyzing oil synthesis by yeast functional complementation experiments .[M ethods] T he yeast expression vector pYES2 .0- P f DGA T1 of P f DGA T1 gene was successfully constructed by double diges-tion ,and the recombinant plasmid was transformed into the yeast mutant strain H 1246 .T he transgenic yeast cells were stained with Nile Red to detect whether oil body could be synthesized in the transgenic yeast .T LC test was used to detect T AG presented in thransgenic yeast .[Results] Oil bodies were able to be detected in the mutant strain H 1246 transfected with pYES2 .0- P f DGA T1 ,but not in the mutant strain H1246 transfected with pYES2 .0 empty vector , w hichindicated that the oil body defectingin Saccharomyces cerev isiae mutant strain H1246was restored by the expres-sion of P f DGA T1 gene.T he diacylglycerolacyltransferase could catalyze T AG synthesis .[Conclusion] T he PfPDA T1 gene plays an important role in the synthesis and accumulation of perilla seed oil .T hese results lay the foundation for the further study of the function of P f DGA T1 gene in the fatty acid metabolism pathway of perilla .关键词
