地熊蜂非视觉相关beta-arrestin基因的克隆与表达谱分析
Cloning and expression spectrum analysis of non-visual beta-arrestin gene in the Bombus terrestris
摘要
Abstract
[Objectives] T he aims of present study were to clone and identify the non-visual beta-arrestin gene of Bombus-terrestris ,to analyze its expression variation at different developmental stages and in specific tissues of queen bees ,and to lay a foundation for the functional research of target gene .[M ethods]PCR amplification and gene cloning were used to obtain the full-length cDNA sequence of the non-visual related beta-arrestin gene.T he sequences of amino acids encoded by the gene were compared and analyzed using bioinformatics techniques .Real-time PCR was used to analyze the gene expression in the brain ,ovary ,midgut ,and fatbody of queens at different developmental stages ,including one-day be-fore pharate stage ,one day after callow stage ,mated for 5-days ,hibernation for 3 months ,hibernation for 5 months , egg-laying stage ,and senescent stage .[Results]T he CDS sequence of beta-arrestin gene was 1248 bp long ,and the size of the encoded protein was 415 aa.T he 27~182 amino acid residues in the amino acid sequence of the protein were deter-mined as the conserved domain of A rrestin-N superfamily ,and the 201~354 amino acid residues were the conserved do-main of A rrestin-C superfamily .T he similarity analysis indicated that the amino acid sequence of the beta-arrestin gene of bumblebee was consistent with that of the transcriptome data collected in NCBI up to 97% .Real-time PCR results showed that beta-arrestin gene was expressed in all developmental stages and tested tissues ,but the expression levels were significantly different .[Conclusion] In this study ,the CDS sequence of bumblebee beta-arrestin gene was cloned ;the amino acid sequence encoded by beta-arrestin gene cDNA was determined ;the sequence characteristics and expres-sion profile of this gene were analyzed ,and it was speculated that it was related to the physiological metabolic process and hormone secretion activity during the queen′s overwinter period .关键词
