Abstract

Objective: To investigate the miR-199a-3p negative regulated pigment box homologous protein 7 (CBX7) on the promotion of the invasion and migration capacity of A549 lung cancer cells ( A549 ). Methods: The expression levels of miR-199a-3p and CBX7 in lung cancer tissues, para-cancer tissues, and lung cancer cell lines were detected by qRT-PCR, and the relationship between miR-199a-3p and clinical characteristics was analyzed. Luciferase reporter assay was performed to detect the binding region of miR-199a-3p with CBX7 mRNA. The effect of overexpression of miR-199a-3p on the expression of CBX7 mRNA in A549 was detected by qRT-PCR. Western blot was used to detect the effect of overex-pression of miR-199a-3p on the expression of CBX7 protein in A549, and to detect the expression of CBX7 protein in lung cancer and paracancer tissues. MTT assay, flow cytometry and Tranwell assay were used to detect the regulation of mir-199a-3p and CBX7 on the proliferation, invasion and migration of A549. Results: The relative expression of miR-199a-3p in lung cancer tissues was significantly higher than that in paracancer tissues. The expression of mir-199a-3p in all lung cancer cell lines was significantly higher than that in normal lung cells, and it was related to TNM stage and metastasis of lung cancer. CBX7 was a possible target gene for miR-199a-3p. MiR-199a-3p could specifically bind to sites in the 3’ UTR region of CBX7 mRNA in a complementary manner, thereby inhibiting the expression of CBX7. Protein and mRNA expression levels of CBX7 in the miR-199a-3p mimics transfected group were significantly lower than those in the control group. The relative expression of CBX7 mRNA and protein in lung cancer tissues was significantly lower than that in paracancer tissues. The ex-pression level of CBX7 mRNA in each lung cancer cell line was significantly lower than that in normal lung cells. MiR-199a-3p inhibited CBX7 and promoted the proliferation, invasion and migration of A549 and the aggregation in the S phase of the cycle. It also down-regulated CBX7 and promoted the proliferation of lung cancer cells. Conclusion: MiR-199a-3p can in-hibit the development of lung cancer by negatively regulating CBX7, and it may be a new target for the diagnosis and treat-ment of lung cancer.
