摘要

Abstract

Objective To investigate the effects of inhibiting TC-1 expression on proliferation, apoptosis, migration and invasion of glioma cells and the underlying mechanisms. Methods Ex-pressions of TC-1 mRNA in glioma U251 cells and normal brain tissue cells were detected by qRT-PCR. The si-NC group ( transfected si-NC ) and the si-TC-1 group ( transfected si-TC-1 ) were transfected into U251 cells by liposome method. Protein expression was detected by Western blotting;cell proliferation by using cell counting kit-8 ( CCK-8) ; apoptosis by flow cytometry; cell migra-tion and invasion by Transwell assay. Results Compared with normal brain tissue cells, the expres-sions of TC-1 mRNA was significantly increased in glioma U251 cells ( P< 0. 05) . In comparison with the si-NC group, the expression of TC-1 mRNA and protein was significantly decreased in the si-TC-1 group, the expression of Bax protein was significantly increased, and the expression of Bcl-2 protein was significantly decreased; cell viability, cell migration and invasion number were signif-icantly decreased, and apoptosis rate was significantly increased ( P < 0. 05) . Conclusion In-hibiting TC-1 expression can inhibit the proliferation, migration, invasion and promote apoptosis of glioma U251 cells. The mechanism may be related to the expression of Bax and Bcl-2 proteins and they might serve as new targets for the prevention and treatment of glioma.
