Effect of YKL-40 gene silencing on the tumor growth of human endometrial carcinoma transplanted in nude mice and tumor CD68 expression

Abstract
Objective:To investigate the effects of YKL-40 gene silencing on the growth of the transplanted tumor and expression of CD68 in tissue of the endometrial carcinoma of nude mice.Methods:We silenced the YKL-40 gene in human endometrial carcinoma (HEC)-1A cells using the small interfering (si)RNA technique and divided the HEC 1A cells into blank control group,mock-treatment group and YKL-40-siRNA group.The HEC-1A cells were injected subcutaneously into the armpits of 4 week-old BALB/c nude mice to establish a nude mouse endometrial cancer model.The growth of transplanted tumors in nude mice was observed.The expressions of YKL-40 and CD68 in tumor tissues were detected by immunohistochemistry.Results:The tumor volume of YKL-40-siRNA transfection group was lower than that of nonsense siRNA transfection group and blank control group.The expression level of YKL-40 was significantly decreased in the tumor tissues of the nude mice in the group subjected to the injection of recombinant lentiviral vectors YKL-40-siRNA (P <0.05).Furthermore,the inhibition of YKL-40 expressiondown-regulated the expression of CD68 in the tumor tissues (P<0.05).Spearman correlation analysis showed that the expression of YKL-40 was positively correlated with the CD68 expression in the tumor tissue of the nude mice.Conclusion:Inhibition of YKL-40 expressionc ould prevent transplanted endometrial carcinoma growth and reduce the expression of CD68.关键词
