Effects of Canine?Derived Compound Probiotics on Cecal Microflora in Splenic Qi Asthenia Canines

Abstract
The objective of this study was to analyze the effects of canine?derived probiotics on cecal microflora in splenic qi asthenia canines using Illumina HiSeq sequencing technology. Twenty?four Beagle canines, aged 1?year?old, were randomly divided into 4 groups, which were control group, splenic qi asthenia group, natural recovery group and probiotic treatment group. The canines in control group were fed a basal diet; the canines in splenic qi asthenia group, natural recovery group and probiotics treatment group were fed the basal diet and were gavaged with 10 mL/( kg BW·d) senna decoction for 7 days to establish splenic qi asthenia model. On the day 7 of trial, the canines in splenic qi asthenia group were dissected for collecting cecal contents; the ca?nines in control group and natural recovery group still continue to fed the basal diet for 7 days; the canines in probiotics treatment group still continue to fed the basal diet and were gavaged with 1×109 CFU/d of canine?de?rived compound probiotic per canine for 7 days. On the day 14 of trial, four canines of each group were ran?domly selected to collect the cecal contents from the control group, the natural recovery group and the probiotic treatment group. The bioinformatics analysis of cecal contents for canines of each group was performed using the Illumina HiSeq sequencing platform and QIIME sequencing software. The results showed as follows: 1) the ACE index, Chao1 index and Shannon index of the cecal microflora in the control group and the probiotics treatment group were higher than those in the splenic qi asthenia group and the natural recovery group, while the Simpson index was lower than that in the splenic qi asthenia group and the natural recovery group. 2) Bac?teroides at different classification levels such as phylum, class, order and family, and bacillus at class and ge?nus levels, their relative abundances in the probiotics treatment group were significantly higher than those in the control group and the splenic qi asthenia group ( P<0.05) , while the relative abundances of other microflora at different classification levels were not significantly different compared with the control group ( P>0.05 ). In summary, canine?derived probiotics can not only restore the cecal microflora diversity in splenic qi asthenia ca?nines to the level of normal canines, but also increase the proportions of beneficial bacteria such as bacteroides and bacillus, and it has a good regulatory effect on the cecal microflora structure of splenic qi asthenia canines.
