Effects of Alfalfa Saponin and Yeast Cultures on Rumen Fermentation Characteristics and Methane Production of Mutton Sheep in Vitro

Abstract
The purpose of this study was to investigate the effects of alfalfa saponin and yeast cultures on rumen fermentation characteristics, methane production and nutrient degradation rate of mutton sheep in vitro. Three healthy Dorper×thin?tailed Han crossbred wethers with an average body weight of ( 67.0 ± 1.3) kg fitted with permanent ruminal fistulas were used for rumen fluid collection. A 3×3 factorial arrangement design was adopt?ed in the experiment, and supplemented with different levels (0, 22 and 44 mg/g DM) of alfalfa saponin and different levels (0, 44 and 88 mg/g DM) of yeast cultures in the fermentation substrate, and carried out by in vitro gas production technique for 48 h in vitro simulated rumen fermentation experiment. The results showed as follows: 1) the interaction effect of alfalfa saponin and yeast cultures had significant effects on cumulative gas production, gas production kinetics parameters, methane production and rumen fermentation fluid parameters in vitro ( P<0.05). 2) With the alfalfa saponin supplemental level increasing, the cumulative gas production and gas production kinetics parameters were linearly increased ( P<0.05) , while the pH and contents of propionic acid and butyric acid in rumen fluid were linearly decreased ( P<0.05) , and the methane production showed a quadratic decrease trend ( P<0.05). 3) With the yeast cultures supplemental level increasing, the gas produc?tion rate, gas production rate when gas production was 1/2 of the maximum and content of total volatile fatty acids in rumen fluid were linearly increased ( P<0.05) , the methane production showed a quadratic decrease trend ( P<0.05) , and the pH in rumen fluid showed linearly and quadratic decrease trend ( P<0.05). 4) The alfalfa saponin and yeast cultures and their interaction effect had no significant effects on nutrient degradation rates in vitro ( P>0.05). In conclusion, the interaction effect of alfalfa saponin and yeast cultures has better effects on rumen fermentation parameters and methane production, and the best combination is 44 mg/g DM alfalfa saponin and 88 mg/g DM yeast cultures.[ Chinese Journal of Animal Nutrition, 2019, 31(9) :4226?4234]关键词
