Abstract

The aim of this experiment was to evaluate the effects of gossypol acetic acid ( GAA) on the num?ber and population diversity of protozoa in rumen fluid of sheep. Eight Kazakh female sheep with permanently rumen fistula were randomly divided into two groups ( control group and trial group) with four sheep in each group. The sheep in the control group were fed a basal diet during trial period ( from 1st to 47th day) , while the sheep in the trial group were fed the basal diet supplemented with 361 mg/kg GAA from 1st to 26th day, and 722 mg/kg GAA from 27th to 47th day of the trial period. Rumen fluid samples at days 20, 37 and 47 were collected to detected the number and population diversity of protozoa by microscope count method com?bined with high throughput sequencing technique. The results showed as follows: GAA significantly reduced the number of protozoa in rumen fluid at day 20 ( P<0.05) , but there was no significant effect at other time points ( days 37 and 47) ( P>0.05). 2) The rarefaction curve of rumen fluid protozoa samples showed that the sequencing had covered most protozoa in rumen fluid. 3) GAA had no significant effect on the alpha diversity of protozoa population in rumen fluid at different time days ( P>0.05). 4) The results of principal component analysis ( PCA) showed that GAA had no obvious effect on the structure of protozoa population in rumen fluid. 5) At genus level, compared with the control group, the relative abundance of Ophryoscolex at day 37 in ru?men fluid of the trial group was significantly increased ( P<0.05) , while the relative abundances of other pro?tozoa genus in rumen fluid were not significantly changed ( P>0.05). In conclusion, under the conditions of this experiment, basal diet supplemented with 361 mg/kg GAA significantly reduces the number of protozoa in rumen fluid of sheep at day 20. There is no significant effect on the number of protozoa in rumen fluid of sheep when increasing GAA to 722 mg/kg. GAA has no significant effect on diversity of protozoa population in ru?men fluid of sheep.[ Chinese Journal of Animal Nutrition, 2019, 31(10) :4582?4589]
