Abstract
Objective To investigate the expression of myelin basic protein (MBP),myelin associated glycoprotein (MAG) and Nogo recepter-interacting protein-1 (Lingo-1) in rat after focal cerebral ischemia-reperfusion injury and to ex-plore the effect of dl-3n-butylphthalide (NBP) on the 3 different proteins. Methods 72 adult rats were divided into sham group,model group and drug group. The animal model of middle cerebral artery occlusion (MCAO) was performed to make cerebral ischemia sustained for 2 h on rats and kept reperfusion for 1 d,3 d,5 d or 7 d. In the drug group,rats received in-traperitoneal injection of NBP with the dose of 20 mg/(kg·d) and the frequency of twice a day. In the sham group,rats re-ceived intraperitoneal injection of saline. Immunohistochemistry was used to detect the expressions of MBP,MAG and Lingo-1 in the rats’ brains. Results In the model group and drug group,the expression of MBP decreases,MAG and Lingo-1 in-creases in rats compared with the sham group. In the drug group,the expression of MBP increases,MAG decreases in rats com-pared with the model group,the differences were statistically significant(P<0. 05). The difference of the expression of Lingo-1 between drug group and model group was not statistically significant (P>0. 05). Conclusion Cerebral ischemia-reperfu-sion can cause the damage on myelin in the cortical area of rats and NBP may have protective effect on impaired myelin.关键词
