Abstract

Objective To analyze the effects of Danhong injection on the anthracyclines-caused cardiotoxicity by regulating Notch signaling pathway. Methods Seventy-five SPF-level Wistar male rats were randomly divided into five groups: normal group, model group, Danhong injection low, medium and high dose groups, 15 rates in each group. Rats in the low, medium and high doses of Danhong injection were injected with Danhong injection at doses of 4 mL·kg-1, 8 mL·kg-1 and 16 mL·kg-1 from the tail vein for 5 days. Except the normal group, rats in the other groups were injected with doxorubicin at a dose of 2.5 mg·kg-1 once a week for 6 weeks, and the cumulative injection dose was 15 mg·kg-1. The general activity, myocardial histopathology, serum oxidative stress and inflammatory factors were observed. The mRNA and protein levels of HIF-1α, Hes1 and Notch1 in myocardial tissue were detected. Results Compared with the normal group, the serum BNP, cTnI and MDA levels increased while the SOD level decreased in the model group and Danhong injection low-dose group (P<0.05). Compared with the model group, the serum BNP, cTnI and MDA levels decreased in the medium and high dose Danhong injection groups, while the SOD level increased (P<0.05). Compared with the normal group, the HIF-1α, Hes1 and Notch1 proteins were upregulated in the myocardial tissue of the model group and low dose Danhong injection group (P<0.05). Compared with the model group, the expression of HIF-1α, Hes1 and Notch1 protein were down regulated in the myocardial tissue of medium and high dose Danhong injection groups (P<0.05). Conclusion Danhong injection can reduce the degree of inflammatory reaction and oxidative stress in myocardial tissue by inhibiting the signal pathway of Notch, and protect the cardiotoxic rats induced by anthracyclines.
