摘要

Abstract

Objective:To investigate the effects of human ribosomal S6 protein kinase 4 mutant 2 (RSK4m2)on proliferation,migration and invasion of breast cancer MDA-MB-231 cells.Methods:MDA-MB-231 cells were transfected with lentivirus vector carrying RSK4m2 (RSK42)to specifically up-regulate the expression of RSK4m2,or transfected with lentivirus negative empty vector as a negative control group.And the untransfected MDA-MB-231 cells were set as a blank control group.The transfection efficiency was confirmed by PCR and western blotting.The proliferation,invasion,and migration of MDA-MB-231 cells were respectively detected by CCK 8,colony formation assay,scratch assay and Transwell assay after transfection.Results:The mRNA and protein expression levels of RSK4rn2 were significantly increased in MDA-MB-231 cells after transfected with RSK4m2.The proliferation,migration and invasion abilities of RSK4m2 group were significantly decreased compared with other two groups(P<0.05).There was no significant difference between the negative control and blank control groups (P>0.05).Conclusion:RSK4m2 is involved in the regulation of biological behavior of human breast cancer MDA-MB-231 cells in vitro,and it can inhibit cell proliferation,migration and invasion.
