Effects of Phospholipid on Astaxanthin Deposition and Utilization of Macrobrachium nipponense

Abstract
The aim of this study was to evaluate the effects of phospholipid on astaxanthin deposition and utili?zation of Macrobrachium nipponense. The phospholipid levels were set at 0, 20 and 40 g/kg, respectively, and the astaxanthin levels were set at 0 and 4 g/kg, respectively. According to the supplemental level of phos?pholipid and astaxanthin, six groups were designed: the control group ( CON group) without phospholipid and astaxanthin supplementation, supplemented 20 and 40 g/kg phospholipid groups ( PL1 and PL2 groups) , sup?plemented 4 g/kg astaxanthin group ( AX group) , supplemented 4 g/kg astaxanthin plus 20 g/kg phospholipid and 4 g/kg astaxanthin plus 40 g/kg phospholipid groups ( AXPL1 and AXPL2 groups). A total of 2 160 Macrobrachium nipponense with initial body weight of (1.00±0.00) g were selected and randomly divided into 3 groups with 3 replicates per group and 120 prawns per replicate. After feeding 4 weeks, twenty prawns were randomly sampled from each replicate and exposed to ( 32.25 ± 0.42 ) mg/L of ammonia concentration in a stress experiment, and sampling and analysis were conducted before and after 24 h of ammonia stress. The re?sults showed that after ammonia stress, the hepatopancreas astaxanthin content of AX, AXLP1 and AXLP2 groups was significantly higher than that of the CON group ( P<0.05) , and AXLP2 group was significantly higher than AX group ( P<0.05). Three?way analysis of variance results showed that dietary phospholipid sig?nificantly increased the astaxanthin content, total superoxide dismutase ( T?SOD) activity and total antioxidant capacity ( T?AOC) in hepatopancreas ( P<0.05) ; dietary astaxanthin significantly increased the hepatopancreas astaxanthin content ( P<0.05) , significantly down?regulated the superoxide dismutase ( SOD) mRNA relative expression ( P<0.05) , and significantly decreased the hepatopancreas T?AOC and T?SOD activity ( P>0.05) ;while ammonia stress significantly increased the astaxanthin content ( P<0.05) , and significantly decreased the hepatopancreas T?AOC and T?SOD activity ( P<0.05). Compared with the CON group, the hepatopancreas B cells number of PL1, PL2, AX, AXPL1 and AXPL2 groups was relative increased. These results suggest that phospholipids improve deposition of exogenous astaxanthin and facilitate utilization of endogenous astaxanthin by Macrobrachium nipponense, and that accumulated astaxanthin in vivo plays an important role in reducing oxidative injury.[ Chinese Journal of Animal Nutrition, 2019, 31(11) :5133?5141]关键词
