Effects of Folic Acid on Liver Oxidative Damage in Lead?Exposed Rats

Abstract
The aim of this study was to investigate the effects of folic acid on lead?induced oxidative stress and nuclear factor E2?related factors ( Nrf2) and heme oxygenase?1 ( HO?1) protein expression in rat liver. Forty 2?month?old male rats were randomly divided into 4 groups ( n=10) and treated as follows: control group, free drinking water+1 mL deionized water was intragastrically administered; lead group, free drink 0.2% lead acetate solution+1 mL deionized water was intragastrically administered; intervention group, free drink 0.2%lead acetate solution+1 mL folic acid suspension was intragastrically administered ( prepared according to the dose of 0.4 mg/kg BW folic acid) ; folic acid group, free drinking water+1 mL folic acid suspension was in?tragastrically administered ( prepared according to the dose of 0.4 mg/kg BW folic acid). Rats in each group were given intragastric administration once a day. After 14 days, the activities of superoxide dismutase ( SOD) , glutathione peroxidase ( GSH?Px) and the contents of malondialdehyde ( MDA) and reduced gluta?thione ( GSH) in rat liver were determined by corresponding kits, and the expression levels of Nrf2 and HO?1 proteins in liver were determined by immunohistochemistry. The results showed that compared with the control group, the liver GSH content and GSH?Px activity in the lead?exposed group were significantly decreased ( P<0.05) , while the serum lead content and the MDA content and the expression levels of HO?1 and Nrf2 proteins in liver were significantly increased ( P<0.05). Compared with the lead?exposed group, the MDA content and the expression levels of Nrf2 and HO?1 proteins in liver of the intervention group were significantly decreased ( P<0.05). Compared with the control group, the serum lead content in the intervention group was significant?ly increased ( P<0.05) , and there were no significant changes in SOD activity, GSH and MDA contents and Nrf2 and HO?1 protein expression levels in liver ( P>0.05).In addition, the activity of GSH?Px in liver of folic acid group was significantly higher than that of the control group ( P<0.05). It is concluded that folic acid has a certain degree of protective effect on liver oxidative damage in lead?exposed rats.[ Chinese Journal of Animal Nutrition, 2019, 31(11) :5178?5183]关键词
