Abstract
Objective To investigate effect mechanism of alantolactone on growth and invasion of retinoblastoma Y79 and phosphorylation of JAK2 STAT3.Methods The retinoblastoma Y79 was cultured in vitro.The cells were divided into 4 groups: Blank control group ( Control), low-dose alantolactone group ( Ala 10 μmol/L ) , medium-dose alantolactone group ( Ala 20 μmol/L ) , high-dose alantolactone group ( Ala 50 μmol/L ) .Cell growth was detected by Edu stai-ning.Apoptosis was detected by Hoechst staining.Cell invasive ability was detected by Transwell chamber experiment.Western Blotting was performed to detect Ki67, PCNA, VEGF, MMP-14 pro-tein expression and phosphorylation of JAK2/STAT3.The mouse model of retinoblastoma was estab-lished.The mice were divided into four groups: Blank control group ( Control), low-dose alanto-lactone group ( Ala 10 μmol/L) , medium-dose alantolactone group ( Ala 30 μmol/L) , high-dose alantolactone group ( Ala 90 μmol/L).The above doses of alantolactone were injected into the ab-dominal cavity of the mice, respectively.The tumor mass was measured 30 days later.The expres-sion of Ki67 and MMP-14 was detected by immunohistochemistry.Results The results of cell ex-periments showed that compared with those in blank control group, after treatment with alantolac-tone, number of cells with red color by retinoblastoma Edu staining, number of cell invasion per u-nit area, Ki67, PCNA, VEGF, MMP-14 p-JAK2 and p-STAT3 were significantly decreased.The above-mentioned indexes were significantly lower in high-dose alantolactone group than in medium-dose alantolactone group, and significantly lower in medium-dose alantolactone group than in low-dose alantolactone group (P<0.05).Compared with those in blank control group, there were no significant changes in protein expression levels of JAK2 or STAT3 in the other groups ( P>0.05) . Compared with that in blank control group, the apoptosis rate was significantly increased.The apop-tosis rate was the the highest in high-dose alantolactone group, followed by medium-dose alantolac-tone group and low-dose alantolactone group ( P<0.05) .The results ofin vivoexperiments of the mice showed that compared with those in blank control group, the tumor mass, positive rates of Ki67 and MMP-14 were significantly decreased after treatment by alantolactone.The above-men-tioned indexes were the highest in low-dose alantolactone group, followed by medium-dose alanto-lactone group and high-dose alantolactone group ( P<0.05).Conclusion Alantolactone can pro-mote apoptosis by decreasing cell invasion and migration related proteins, increasing apoptosis-relat-ed proteins, and inhibiting invasion and migration of retinoblastoma Y79.关键词
